The next generation of RNA vaccines: self-amplifying RNA

چکیده
The global COVID-19 pandemic has brought tremendous momentum to the field of messenger RNA (mRNA) vaccines. advantages this vaccine platform, such as rapid development and high efficacy, resulted in mRNA vaccines being first approved against COVID-19. Looking forward future vaccines, how can we make even better? While improvements stability formulation cost are inevitable, one main challenges is lowering dose order avoid side effects associated with doses RNA. One way do by using self-amplifying (saRNA), a type that encodes replicase copies original strand once it’s cell. Here, discuss origins saRNA, it works comparison mRNA, current saRNA

Nonviral delivery of self-amplifying RNA vaccines.
Despite more than two decades of research and development on nucleic acid vaccines, there is still no commercial product for human use. Taking advantage of the recent innovations in systemic delivery of short interfering RNA (siRNA) using lipid nanoparticles (LNPs), we developed a self-amplifying RNA vaccine. Here we show that nonviral delivery of a 9-kb self-amplifying RNA encapsulated within ...
متن کاملDevelopment of nucleic acid vaccines: use of self-amplifying RNA in lipid nanoparticles
Self-amplifying RNA or RNA replicon is a form of nucleic acid-based vaccine derived from either positive-strand or negative-strand RNA viruses. The gene sequences encoding structural proteins in these RNA viruses are replaced by mRNA encoding antigens of interest as well as by RNA polymerase for replication and transcription. This kind of vaccine has been successfully assayed with many differen...
متن کاملRNA Sequencing with Next-Generation Sequencing
Sequencing of RNA has been an important application of DNA sequencing technology since its invention. RNA is usually sequenced by first converting it to complementary DNA (cDNA) with the reverse transcriptase enzyme (RNA-dependent DNA polymerase). Reverse transcriptase was originally isolated from Rous sarcoma retrovirus and Rauscher mouse leukemia retrovirus (R-MLV) by Baltimore (1970) and ind...

متن کاملEnhancement of RNA Interference Effect in P19 EC Cells by an RNA-dependent RNA Polymerase
Background: RNA interference (RNAi) is a phenomenon uses double-stranded RNA (dsRNA) to specifically inhibit gene expression. The non-specific silencing caused by interferon response to dsRNA in mammalian cells limits the potential of utilizing RNAi to study gene function. Duplexes of 21-nucleotide short interfering dsRNA (siRNA) inhibit gene expression by RNAi. In some organisms, siRNA can als...
